Base editing is back. Here we look at how base editing was employed in a recent study to repair the genetic mutation that causes Hutchinson-Gilford progeria syndrome (progeria) in a mouse model. Progeria causes accelerated aging and patients with this disease typically die around the age of 14 or 15 years. Remarkably, with a single AAV injection of an adenine base editor and a gRNA targeting the Lamin A gene (LMNA) that is mutated in progeria, the median lifespan on the mice were more than doubled!
